Introducing pOET3: A New Vector for Improved Post Translational Modification of Your Protein
OET is delighted to announce the launch of a new transfer vector, pOET3. With the launch of this product you can harness the following capabilities to improve your protein expression… · High level expression of foreign genes under the late AcMNPV basic (p6.9) promoter. · Earlier expression compared to the polyhedrin promoter. · Small size to facilitate cloning · Compatible with all baculovirus systems based on homologous recombination · Enhanced expression of proteins that require extensive posttranslational modifications such as glycosylation. When combined with the capabilities of the flashBAC range for improved protein quality and yield this can help improve your recombinant protein production still further. The launch of pOET3 is complemented by the launch of pOET2. pOET2 offers the same multiple cloning site as pOET1 but in reverse orientation offering more flexibility at the cloning stages of your work.

New Transfer Vector Now Available: pOET-1TM
pOET-1TM is a baculovirus transfer vector designed for high levelexpression of foreign genes under the powerful AcMNPV polyhedrin (polh) promoter.pOET-1TM has a Col E1 origin of replication and an Ampicillin resistancegene for selection in E. coli. The vector is smaller than other available transfervectors (4541 bp) which greatly facilitates the cloning steps. The polh sequenceshave been replaced by a multiple cloning site containing 14 unique restrictionsites for insertion of the foreign gene in the correct orientation. The PacIsite at the end of the MCS provides translational stop codons in all threereading frames for expression of truncated proteins. The AcMNPV sequences flankingthe gene in the transfer vectors MCS allow recombination with the viral DNAto insert the expression cassette into the polh locus. As well as our own flashBACTM platformpOET-1TM can also be used with any other baculovirus system thatuses homologous recombination in insect cells. Moreinformation is available on this web site.

Virus titres in two hours – and no plaque assayrequired!
The baculoQUANT kitis the perfect companion to the flashBAC rangeand is an essential tool in any laboratory working with baculoviruses. baculoQUANT isa novel, one step, virus titration kit that gives an accurate result withintwo hours without the need for plaque assays or immunoassays. BaculoQUANT isbased on quantitative PCR (qPCR) technology. QPCR utilizes a set of primersand a probe which are designed to specifically target and amplify a small regionof the baculovirus genome. The increase in amplification of this small regionis then correlated to virus genome copy number and can be used to measure thenumber of recombinant virus particles present in an infected insect cell culture.This novel method has been proven to be a very accurate, reproducible and fastway to titrate recombinant baculoviruses1, both for single-use titrations andfor high throughput applications on automated systems.
